Hydroxycarboxylic acid receptors in GtoPdb v.2023.1

چکیده

The hydroxycarboxylic acid family of receptors (ENSFM00500000271913, nomenclature as agreed by the NC-IUPHAR Subcommittee on Hydroxycarboxylic [36, 12]) respond to organic acids, including endogenous hydroxy carboxylic acids 3-hydroxy butyric and L-lactic acid, well lipid lowering agents nicotinic (niacin), acipimox acifran [53, 60, 65]. These were provisionally described receptors, although shows submicromolar potency at HCA2 only is unlikely be natural ligand [60,

Hydroxycarboxylic acid receptors are essential for breast cancer cells to control their lipid/fatty acid metabolism

Cancer cells exhibit characteristic changes in their metabolism with efforts being made to address them therapeutically. However, targeting metabolic enzymes as such is a major challenge due to their essentiality for normal proliferating cells. The most successful pharmaceutical targets are G protein-coupled receptors (GPCRs), with more than 40% of all currently available drugs acting through t...

Supramolecular aggregation of two hydroxycarboxylic acid derivatives.

The crystal structures of 7,7-dicyclobutyl-5-hydroxymethyl-6-oxabicyclo[3.2.1]octane-1-carboxylic acid, C17H26O4, (I), and 1-(hydroxymethyl)-7-oxaspiro[bicyclo[3.2.1]octane-6,1'-cyclopentane]-5-carboxylic acid, C13H20O4, (II), determined at 170 K, show that the conformation of the hydroxymethyl group (anti or gauche) affects the dimensionality (one- or two-dimensional) of the supramolecular str...

3,5-Dihydroxybenzoic acid, a specific agonist for hydroxycarboxylic acid 1, inhibits lipolysis in adipocytes.

Niacin raises high-density lipoprotein and lowers low-density lipoprotein through the activation of the β-hydroxybutyrate receptor hydroxycarboxylic acid 2 (HCA2) (aka GPR109a) but with an unwanted side effect of cutaneous flushing caused by vascular dilation because of the stimulation of HCA2 receptors in Langerhans cells in skin. HCA1 (aka GPR81), predominantly expressed in adipocytes, was re...

Hydroxycarboxylic acid receptor 2 mediates dimethyl fumarate's protective effect in EAE.

Taken orally, the drug dimethyl fumarate (DMF) has been shown to improve functional outcomes for patients with MS; however, it is unclear how DMF mediates a protective effect. DMF and, more so, its active metabolite, monomethyl fumarate, are known agonists of the hydroxycarboxylic acid receptor 2 (HCA₂), a G protein-coupled membrane receptor. Here, we evaluated the contribution of HCA₂ in media...

Highly stereoselective synthesis of α-alkyl-α-hydroxycarboxylic acid derivatives catalyzed by a dinuclear zinc complex.

A dinuclear zinc-ProPhenol catalyst enables highly enantioselective nitro-Michael reactions with oxazol-4(5H)-ones as nucleophilic substrates (see scheme, Nap = 2-naphthyl). This work highlights the utility of the ProPhenol family of ligands. The modular nature of these ligands proved crucial in the optimization of reaction conditions to achieve excellent stereoselectivities.
